Passage 1 Neonatal Rat Cardiac Fibroblastscell can be shipped proliferated in a T-25 flask at >90 confluence, or frozen vial (> 5x 105 cells). DMEM contains 5% Fetal calf serum (Full medium) is recommended forcell culture and these cells have an average minimum population doubling levels >8when cultured following the detailed protocol. Cells are tested negative for commonexperimental animal pathogens, and mycoplasma in vitro. When you receive thecells, leave the flask in 37°C CO2 incubator for 1 hour first, and then replace thetransport medium with fresh Full medium. Let the cells grow for 24hours beforesubculture.Subculturing Proliferated Cells:1. Rinse the cells in T25 flask with 5ml DPBS (Room Temperature, RT) twice.2. Add 2ml of Trypsin/EDTA (RT) into T25flask (make sure the whole surface ofthe T25 flask is covered with Trypsin/EDTA),and gently dispose theTrypsin/EDTA solution within 10 seconds with aspiration.3. Leave the T25 flask with the cells at RT for 1 minute (the cells will normallycome off the surface within 1-2 minute). Suspend the cells with 40ml of freshFull medium and the cell suspension is transferred directly into 4 x T25 flasks(10ml each, and the cells are subcultured at 1:4ratio).4. Culture medium (full medium) is changed every 2-3 days. The cells normallybecome confluent within 7 days (when split with a ratio of 1:4).
