_____________________________________________________________________General Information: The retina is an important in vitro model for the central nervoussystem. It is more readily accessible than most nervous tissue and is strong enough toremain intact during manipulations. The retina of the rabbit commonly contains nopenetrating blood vessels. It is nurtured by diffusion from capillary networks on eitherside. For this reason, the tissue will survive and functions without circulating blood, aslong as it is bathed in a medium that closely resembles in vivo conditions.Ames’ medium was formulated to support retinal tissue in relatively short-term culture.Rabbit retina has been incubated in Ames’ medium for over 2 days with its metabolismand electrical responses to light stimuli well maintained. This mixture is a medium ofchoice for maintaining central nervous system tissue in vitro.The medium is for research use only and not approved for human or animal use or forapplication in any in vitro diagnostic procedure. Storage of the medium andsupplements Please note that the basal medium has to be stored at 4°C up to 8°C butnever should be frozen. The supplement has to be stored at -20°C. Protect from light fora longer time period.Preparing and Stability:Thaw the supplement mixture at 37°C before adding to the basal medium. After thawingplease rinse the bottle and the tube with 70% ethanol as protection. Remove ethanolexcess before opening the cap of the tube. After adding the supplement mixture to thebasal medium slew carefully to get a homogenous medium. Please be careful not to touchthe tube or bottle interior threads with your fingers. Please avoid long term exposure tolight because some components are light sensitive. Also do not warm up the medium over37°C. All this protects your medium against contamination or degradation of essentialcomponents.
